چکیده

It has been claimed that, in patients with amyloidosis, the presence of amyloid material in the skin may be demonstrated by the intracutaneous injection of Congo red (Philpott and Freshman, 1936; Nomland, 1936). In the presence of amyloidosis, the area of staining is said to be more intense and to persist for a longer time than in normal skin. Recently this technique has been employed by Laine, Holopainen, and Makinen (1956) in a series of patients with rheumatoid arthritis. The fixation of the dye in the skin was measured by the time taken for the stained area to fade to a standard intensity and this was found to be significantly longer in 55 patients with rheumatoid arthritis than in thirty normal control subjects. From these results, Laine and his co-workers concluded that this phenomenon might be due to the presence of amyloid in the skin of patients with rheumatoid arthritis. This seemed quite possible since, with the decline of tuberculosis, rheumatoid arthritis is one of the chief causes of secondary amyloid. At the Postgraduate School of Medicine, London, amyloid was found in eight out of 48 autopsied cases (Missen and Taylor, 1956). Using a modified method of assessment, we have confirmed Laine's results, but our experience leads us to differ somewhat from Laine and his colleagues in their tentative conclusions cited above.

Resorption of intracutaneously injected Congo red.

Since the introduction of the intravenous Congo red test by Bennhold (1923), a number of writers have recorded observations on the intracutaneous injection of Congo red dye. Philpott and Freshman (1936) and Nomland (1936) observed intense and prolonged staining of cutaneous amyloid deposits in patients following the local injection of Congo red solution. Laine, Holopainen, and Makinen (1956) no...

Cutaneous manifestations in patients with rheumatoid arthritis

Rheumatoid arthritis (RA) is a chronic inflammatory joint disease with different extra-articular manifestations. Among those extra-articular manifestations, there are significant cutaneous manifestations which may result in first admission to a dermatologist. In this article some specific and non-specific skin lesions of RA such as rheumatoid nodules, exacerbated nodules, rheumatoid vascu...

Relation between the affinity for congo red and the glycoprotein content of serum in rheumatoid arthritis and related diseases.

Disappearance of congo red from the serum has long been considered by many investigators to be the best and most constant indicator of amyloidosis. The congo red test, as shown by Bennhold (1923), measures the proportion of this dye which is retained in the plasma one hour after its intravenous injection. The validity of this test has been the subject of intense discussion. Disappearance of 40-...

Lymph node hyalinisation in rheumatoid arthritis and systemic sclerosis.

AIMS To review the histological features of lymph nodes excised from seven patients with rheumatoid arthritis and one with systemic sclerosis. METHODS Lymph nodes excised from seven patients with rheumatoid arthritis and one patient with systemic sclerosis over a 10 year period were examined using the stains haematoxylin and eosin, periodic acid Schiff (PAS), Masson-trichrome, and Congo red f...

A quantitative method for detecting deposits of amyloid A protein in aspirated fat tissue of patients with arthritis.

OBJECTIVE To describe a new, quantitative, and reproducible method for detecting deposits of amyloid A protein in aspirated fat tissue and to compare it with smears stained with Congo red. METHODS After extraction of at least 30 mg of abdominal fat tissue in guanidine, the amyloid A protein concentration was measured by a monoclonal antibody-based sandwich ELISA. RESULTS The concentrations ...
